Abstract
Objective:The Network pharmacology and Drug Component-Target Molecular Docking Technology were used to study the molecular mechanism of knee osteoarthritis (KOA) in the treatment of knee osteoarthritis (KOA), so as to provide theoretical support for clinical application. Methods: Firstly, the main active ingredients and targets in the decoction were retrieved through the TCMSP database, and the known targets of the unobtained active compounds were predicted according to the published literature reports and Swiss Target Prediction. The relevant targets of KOA were retrieved through GeneCards, DisGeNET, OMIM and TTD databases, and the intersection targets of knee three visceral decoction and KOA were obtained by using the Wayne diagram. Subsequently, the drug-disease-target network diagram and PPI network were constructed by the STRING11.0 database combined with Cytoscape3.9.1 software, and the core components and targets of the treatment of KOA were screened. Results: A total of 79 drug-disease intersection targets were screened, with luteolin, β-dermatone and baicalein as the core compounds of drug action, and PTGS2, ESR1, ESR2 and CDK2 as the core targets. Molecular docking verification showed that PIK3CA had the best binding activity with β-desmatone. The mechanism of the treatment of KOA mainly involves the mitogen-activated protein kinase signaling pathway and the nuclear factor-activated B cell kappa-light chain enhancer signaling pathway. Conclusion Knee three organs decoction can treat osteoarthritis through multi-component, multiple target categories as well as multiple pathways, which provides ideas and theoretical basis for further research and experimental work.
